Quantitative and dynamic cell polarity tracking in plant cells

Quantitative information on the spatiotemporal distribution of polarised proteins is central for understanding cell-fate determination, yet collecting sufficient data statistical analysis difficult to accomplish with manual measurements. Here we present Polarity Measurement (Pome), a semi-automated pipeline quantification cell polarity and demonstrate its application variety developmental contexts. Pome reveals that, during asymmetric divisions in Arabidopsis thaliana stomatal lineage, BASL BRXL2 are more asynchronous less mutually dependent than previously thought. A similar linearly arrayed lineage Brachypodium distachyon revealed that MAPKKK BdYDA1 segregated following asymmetrical divisions. Our results versatile tool, which by itself or combined tissue-level studies advanced microscopy techniques can help uncover new mechanisms polarity.
